Frequent micro-awakenings during sleep could be an early indicator of Alzheimer's disease long before memory problems first appear, researchers revealed in a study published in Sleep.

Scientists analyzed sleep data from more than 500 healthy volunteers to determine whether subtle night disruptions correlate with genetic susceptibility to cognitive decline. Each participant received a polygenic risk score for Alzheimer's disease calculated from their genetic profiles.

A polygenic risk score estimates an individual's hereditary likelihood of developing a specific condition by aggregating the effects of thousands of genetic variants. In neurodegenerative research, polygenic profiling helps scientists identify high-risk individuals decades before cognitive decline manifests.

The study found that adults aged 50 to 69 who carried a higher genetic risk experienced a significantly greater frequency of night micro-awakenings. These events are defined as brief bursts of brain activation that usually do not wake a person up completely, but they disturb the overall structure of sleep. No such connection was identified among younger study participants.

Micro-awakenings are transient disruptions lasting only a few seconds that alter brain wave activity during sleep. Although affected individuals rarely remember waking up, these micro-arousals interrupt normal sleep architecture, preventing the brain from completing critical non-REM and REM sleep cycles necessary for cellular maintenance and memory consolidation.

Brainstem Structure and Early Protein Deposits

To understand the biological mechanisms behind the link, researchers focused on the locus coeruleus, a small area in the brainstem roughly the size of a grain of rice. This anatomical structure is responsible for regulating sleep and wakefulness cycles.

Medical experts identify the locus coeruleus as one of the primary sites where pathological proteins first accumulate in the early development of Alzheimer's disease. According to the study authors, nighttime disruptions may be driven by functional changes in this region long before any clinical symptoms become noticeable.

Alzheimer's disease is a progressive neurological disorder that damages nerve cells in the brain, leading to memory loss and cognitive decline. It is the most frequent cause of dementia worldwide, typically affecting older adults as abnormal protein deposits disrupt brain communication networks.

Future Screening and Diagnostic Potential

The researchers emphasized that the current findings represent a statistical association rather than a practical diagnostic test for the disease. However, they pointed out that routine sleep monitoring could eventually serve as a simple, non-invasive tool to identify individuals at elevated risk for Alzheimer's disease.

The study builds on a growing body of scientific research examining early risk factors for cognitive impairment. In prior findings, scientists established that restricting sugar consumption during a child's first 1,000 days of life reduces the long-term risk of developing Alzheimer's disease.

The research paper appeared in Sleep, a peer-reviewed academic journal published by Oxford University Press on behalf of the Sleep Research Society. The publication specializes in clinical sleep medicine, neurobiology, and sleep disorder research.
