The popular medication semaglutide extended the lifespan of elderly healthy mice and weakened several age-related bodily changes, according to research published in the scientific journal Nature.

Researchers administered semaglutide to 20-month-old female mice during a three-month trial. Following the treatment period, the animals showed measurable improvements in both muscle function and cognitive abilities.

An analysis of gene activity after three months of treatment revealed a weakening of several characteristic signs of aging. The authors reported reductions in chronic low-grade inflammation and an improvement in the body's capacity for tissue regeneration.

Semaglutide is a pharmaceutical drug classified as a glucagon-like peptide-1 receptor agonist, widely prescribed for the treatment of type 2 diabetes and long-term weight management. The active ingredient mimics GLP-1, a naturally occurring gut hormone that stimulates insulin secretion, slows stomach emptying, and signals feelings of fullness to the central nervous system.

In laboratory research, mice are widely used to study mammalian aging because their genetics and cellular processes closely resemble those of humans. Laboratory mice typically live between two and three years, making 20-month-old subjects representative of an advanced age stage appropriate for testing late-life health interventions.

Lifespan Extension and Calorie Comparison

In a separate experiment described in the study, mice received semaglutide continuously for the remainder of their natural lives. The median lifespan of the animals receiving semaglutide was nearly 100 days longer than that of mice in the untreated control group.

Scientists also compared the effects of semaglutide against a dietary regimen with a 24 percent reduction in caloric intake. They found that the drug reproduced many of the biological benefits associated with calorie-restricted diets.

On specific health metrics, including spatial memory performance and blood sugar control, semaglutide demonstrated additional advantages compared to the 24 percent caloric restriction diet alone.

Caloric restriction without malnutrition has long been studied in gerontology as one of the most reliable non-pharmacological methods for delaying age-related decline in animal models. Spatial memory, which involves the brain's ability to record and navigate spatial surroundings, is among the cognitive functions that typically deteriorate as organisms age.

Biological Mechanisms and Trial Limitations

The authors suggested that semaglutide may act on biological mechanisms of aging through pathways beyond the simple reduction of food intake. The findings indicate that the pharmaceutical compound may trigger underlying cellular protective effects independently of reduced appetite.

However, the researchers emphasized that the experiments were conducted exclusively on elderly female mice. Consequently, the authors cautioned that the results cannot be directly transferred or applied to humans.

The research joins a growing body of scientific inquiry into how established pharmaceutical treatments may offer broader health benefits in older age. Scientists previously established that statin medications lower the risk of heart attack and stroke by 30 percent in individuals over the age of 70.

Statins are a class of lipid-lowering drugs that inhibit HMG-CoA reductase, an enzyme essential for cholesterol synthesis in the liver. They are widely prescribed to reduce low-density lipoprotein cholesterol and lower the risk of cardiovascular events in aging populations.
